DNA barcoding is a modern method for identification of different species including insects. Amonganimals, the major emphasis about DNA barcoding is on insects. Due to this global trend we addressed herethis approach for surveying on a group of insects and their natural enemies. The aphids of pome fruitorchards and their parasitic wasps (including primary and secondary parasitoids) were collected from Mashhadcity through 2009-2010. Preliminary identification of this group was performed by using morphological,morphometric characters and SEM which finally confirmed by related specialist. Afterward, COI gene inthose specimens was amplified and sequenced. Inroduced species of aphids are Aphis pomi, Allocotaphisquaestionis, Dysaphis affinis, D. plantaginea and Nearctaphis backeri. Collected parasitoids were Aphidiusmatricariae, Binodoxys angelicae, Diaeretiella rapae, Ephedrus persicae, Lysiphlebus fabarum and Praonvolucre and hyperparasitoids species were Alloxysta sp. , Asaphes suspensus, Dendrocerus carpenteri,Pachyneuron aphidis and Syrphophagus aphidivorus. Based on intra-interspecies distances and phylogeneticanalysis using NJ, all species possess diagnostic barcode sequences. The results of this study indicated thatCOI sequence could be useful in species delimitation of these insect groups. Here we provided first genbankdata for COI gene of above mentioned hyperparasitoids plus an initial attempt toward preparing DNAbarcodes for Iranian aphids, their parasitoids and hyperparasitoids
